It's just depending on what you are.
If you are a dog, you can first mark the area by urinating on the boundaries. Then you can proceed to bite anyone that comes into your area.
If you are a lizard, you leave your scent around the boundaries. Then you can either loose your tail or bite off tails of others.
If you are an army general, you plan your attack and conquer the area. Then you build army posts around the boundaries to defend it.
If you are an opportunistic developer, you come in the middle of the night when everyone is sleeping and push down the whole area flattening everything. Then you can build signs saying you will shoot anyone coming into your area.
